PROCEDURE FOR HB ESTIMATON BY AUTOMATION
1. Blood is aspirated from the sample tube into the machine.
2. 6 µl of blood is transferred to the transducer chamber along with 1.994ml of diluent. At the same time 1.0ml of Hb lyse regent is added to prepare 1:500 dilution sample, which, in 10 seconds start reacting making RBCs haemolysed and platelet shrink, there for at the same time Hb is converted into red colored methaemoglobin.
3. Then about 1.0ml of the solution is transferred to the Hb flow cell.
4. In Hb flow cell 555-nm wavelength beam irradiated from the light immitting diode (LED) is applied to sample.
5. Concentration of his sample is measured as absorbance.
6. This absorbance is compared with that of the diluents (the Hb lyse reagent) which was measured before addition of the sample, there by calculating the Hb value (CDC, 2005)
CALCULATION OF HAEMATOLOGICAL INDICES
RBCs constant are the RBCs indices: Mean RBCs Volume, Mean RBCs Haemoglobin and Mean RBCs Haemoglobin concentration.
They are calculated from Hb, RBCs and HCT as follows
