    • 3.4.2 Determination of fungi count
      Plating was done on Sabouraud Dextrose Agar using pour plate method. Serial dilution was carried out using 1ml of each of local drinks samples to 9ml of normal saline to reduce the microbial load. After dilution, 1ml was then poured into Petridis before the addition of cooling sabouraud dextrose agar on the plates. The plates were swirled gently and allowed to solidify. They were then incubated at room temperature 280C for 48-96 hours.
      3.4.3 Identification of Fungi isolate:
      Identification of fungi isolate were carried out by adding a drop of lacto phenol cotton blue stain placed on a clean grease free glass slide and a sterile inoculating needle used to tease out a fragment of the fungi and transferred into the stain on the slide and covered with a cover slip and examined with low power objective of the microscope x40 objective lens.
       3.5 Germ Tube Test
      Germ tube test was carried out on the fungi isolated to identify Candida specie.
      Materials: Culture suspected Candida colonies from cultured plate.
      Reagent serum( human or sheep red cell), test tube, loop or wooden applicator pasture pipette, slides, cover slips).
      Procedure: 0.5ml of serum was added into a clean test tube, a light suspension of the sample colony were made by touching 1to 2 or 3 colonies using a sterile straight wire loop on the serum. The tube were incubated for 2-3hours at 35-30C. A drop of the suspension were placed on a slide with the aid of a sterile Pasteur pipette and then covered with cover slip.The preparation were examined for production of germ tubes using x10 first and then x40 objective lenses.
      Result:Long tube-like projection extending out from the yeast cell (A positive result shows a hyphae (filamentous) extension arising laterally from a yeast cell with no constriction at the point of origin may be confirmed as Candida albicans. No production of tube-like projection with no hyphae confirmed negative Germ Tube Test (GTT).
