    • 3.8    CALCULATION OF RESULTS
      1. The mean absorbance value (A450) for each set of reference standards and samples was calculated.
      2. A standard curve was constructed by plotting the mean absorbance obtained for each reference standard against its concentration in pg/ml on a linear-linear graph paper, with absorbance values on the vertical or Y axis, and concentrations on the horizontal or X axis.
      3. The mean absorbance values for each specimen was used to determine the corresponding concentration of Estrogen in pg/ml from the standard curve.
      3.8.1 DETERMINATION OF PLASMA FASTING GLUCOSE
       METHOD:    Glucose Oxidase Peroxidase (GOD-POD) according to Trinder, (1969).
      PRINCIPLE:
      Enzymatic colorimetric determination of glucose according to the following reaction.

      Glucose is oxidized by Glucose oxidase and produces gluconate and hydrogen peroxide. The hydrogen peroxide is oxidatively coupled with 4-aminoantipyrine and phenol. The intensity of the coloured complex (quinoneimine) is proportional to the glucose concentration in the sample and can be measured photometrically at 505nm.  
      Procedure for Glucose estimation:
      i.    Clean test tubes were labelled as blank standard and sample
      ii.    20 µL    of test and standard was dispensed into the labelled test tubes respectively.
      iii.    2000 µL (2ml) of glucose working reagent was added to all the tubes.
      iv.    It was mixed and incubated for 20minutes at room temperature
      v.    Absorbance was read spectrophotometrically at 505nm against reagent blank.
